Description |
Caspofungin Acetate is a pioneering antifungal agent belonging to the echinocandin class. It is used intravenously for the treatment of invasive aspergillosis in patients who are refractory to or intolerant of other antifungal therapies. Caspofungin is a stable, water-soluble derivative of pneumocandin B, a fermentation product of the fungus Zalerion arboricola. It stands out by inhibiting the synthesis of 1,3-beta-D-glucans present exclusively in the fungal cell wall, leading to specific lysis of pathogenic cells. This fungicidal characteristic is unusual among antifungal agents. |

With a focus on fungus-specific targets, Caspofungin demonstrates efficacy against various Candida species, including those resistant to azoles and amphotericin B. It also acts against Aspergillus, a significant pathogen resistant to fluconazole. However, Cryptococcus is not susceptible to Caspofungin. Unlike azoles, Caspofungin does not inhibit cytochrome P450. The drug, given intravenously with a loading dose of 70 mg followed by 50 mg daily, has proven effective in treating invasive aspergillosis refractory to prior antifungal therapy. Caspofungin has a human half-life of around 10 hours, and its side effects are generally mild, including fever, thrombophlebitis, headache, nausea, rash, and flushing. As an echinocandin, it plays a crucial role in inhibiting β (1,3)-D-glucan synthesis, an integral component of the fungal cell wall, making it a valuable antifungal drug for the treatment of invasive fungal infections. |
